Primary anti-cancer mechanisms of silymarin and milk thistle

Silymarin is the active extract of Milk Thistle, so the mechanisms below describe the biological actions of Milk Thistle / Silymarin / Silibinin in oncology research.

Overview

Silymarin / Milk Thistle works across multiple cancer-relevant pathways at once. That broad activity is one reason it remains of interest as a pleiotropic adjunct rather than a single-target compound.

1. Apoptosis induction

  • Activates both intrinsic and extrinsic apoptotic pathways

  • Shifts Bcl-2/Bax balance toward cell death

  • Increases caspase activation and PARP-related apoptotic signalling

2. Cell-cycle arrest

  • Frequently induces G0/G1 arrest

  • Slows replication and proliferation in multiple cancer cell lines

3. Anti-proliferative signalling

  • Inhibits EGFR and downstream ERK1/2 signalling

  • Reduces cyclin D1, survivin, c-MYC, and related growth signals

  • Interferes with PI3K-linked survival pathways in selected models

4. Anti-angiogenic effects
